چکیده

Backgr ound and Objective: To examine the acute effects of propylthiouracil (PTU) on corticosterone release by rat adrenal Zona Fasciculata Reticularis (ZFR) cells. Materials and Methods: Male rats were infused with alkaline saline or PTU (10 mg kgG1) via right jugular catheter. The blood samples were collected at 0, 30, 60, 120, 180 and 240 min after infusion. Rat ZFR cells were treated with PTU (12 mM) in the presence or absence of adrenocorticotropic hormone (ACTH) (10G9 M). Media were collected for corticosterone RIA and cells were lysed for analyzing the protein expressions of Steroidogenic Acute Regulatory (StAR) protein and P450scc. The ZFR cells were also incubated with PTU (3-30 mM) plus trilostane (a 3ß-hydroxysteroid enzyme inhibitor, 10G6 M) or both trilostane and 25-OH-cholesterol (10G7~10G4 M) for 1 h and the concentration of pregnenolone in media was measured. Results: Propylthiouracil infusion decreased plasma corticosterone concentrations. Both basal and ACTH-stimulated corticosterone release as well as StAR protein expression in ZFR cells were attenuated by PTU treatment. The Electrophoretic Mobility Shift Assay (EMSA) results showed that the binding activity into Steroidogenic Factor-1 (SF-1) binding site was descreased by PTU. The elevated binding activity in response to ACTH treatment was diminished after PTU treatment. The pregnenolone production from ZFR cells was suppressed by PTU. Administration of PTU (10 mM) decreased Vmax by 46%. Conclusion: Propylthiouracil acutely diminished corticosterone production by affecting the early rate-limiting steps of steroidogenesis including the reduction of the enzyme activity of P450scc, and the expression of StAR protein.

The role of neuromedin B in the regulation of rat pituitary-adrenocortical function.

The effects of a 7-day administration of neuromedin B (NMB) and/or (Tyr4, D-Phe12)-bombesin, an NMB-receptor antagonist (NMB-A) on the function of pituitary-adrenocortical axis were investigated in the rat. NMB raised the plasma concentration of aldosterone, without affecting that of ACTH or corticosterone; the simultaneous administration of NMB-A prevented the effect of NMB. Neither NMB nor NM...

Relation of intracellular K+ and steroidogenesis in isolated adrenal zona glomerulosa and fasciculata cells.

1. Intracellular K+ content, water spaces and corticosterone output were measured in isolated zona glomerulosa and zona fasciculata-reticularis cell suspensions of rat adrenal cortex, after incubation in vitro under conditions designed to alter steroidogenesis. 2. Intracellular K+ of unpurified zona glomerulosa cells was not altered after stimulation of corticosterone output with serotonin. Sim...

Effects of neuromedin-U on immature rat adrenocortical cells: in vitro and in vivo studies.

Neuromedin U (NMU) is a brain-gut peptide, that in the peripheral organs and tissues acts via a G protein-coupled receptor, called NMUR1. Reverse transcription-polymerase chain reaction showed the expression of NMUR1 mRNA in either cortex and medulla or dispersed zona glomerulosa and zona fasciculata-reticularis cells of the immature rat adrenals. Accordingly, immunocytochemistry demonstrated t...

An Oxidized Metabolite of Linoleic Acid Stimulates Corticosterone

Oxidized derivatives of linoleic acid have the potential to alter steroidogenesis. One such derivative is 12,13-epoxy-9-keto-10(trans)-octadecenoic acid (EKODE). In order to evaluate the effect of EKODE on corticosterone production, dispersed rat zona fasciculata/reticularis (subcapsular) cells were incubated for 2 h with EKODE alone, or together with rat ACTH (0, 0.2 or 2.0 ng/ml). In the abse...

Accumulation of steroidogenic acute regulatory protein mRNA, and decrease in the secretory and proliferative activity of rat adrenocortical cells in the presence of proteasome inhibitors.

Sporadic findings indicate that proteolysis may affect steroid secretion in rat ovary granulosa cells. We examined the effects of the proteasome inhibitors MG115 and MG101 on the in vitro secretion and growth of rat adrenocortical cells. MG115 and/or MG101 decreased within 120 min the aldosterone and corticosterone secretion from freshly dispersed zona glomerulosa and zona fasciculata-reticular...
